陈瑾坦言饰演“敦煌女儿”樊锦诗让自己压力山大，几度想要放弃，“越了解，越紧张，她对于我来说不仅是一个角色，更是一个与高尚灵魂不断接近的过程。”她也希望每个女性都能像樊老一样找到自己热爱的事业，自信地绽放。
电影中，樊锦诗与丈夫彭金章相知相守数十年的“神仙爱情”令观众动容。饰演彭金章的演员黄品沅这样解读：“他们两个人能够在那样艰苦的环境里扎根大西北，相互扶持，相濡以沫几十年没有大爱是做不到的，而这种爱就是对敦煌这座世界艺术文化宝库的大爱。”
导演苗月向现场观众分享了《吾爱敦煌》的艰难创作过程。电影从筹备至电影上映前后历时五年，在取材、采风勘景，探访原型人物等前期工作中就下足功夫，五年的筹备也让主创们感受到创作历史传记电影之不易。但为了呈现伟大女性的人生事迹，再艰难也毅然决定坚持往下走。
在拍摄过程中遇到的最大困难就是如何在保护文物的同时保证电影画面质量，为此剧组与敦煌研究院进行了大量的沟通，利用模拟窟拍摄，利用镜子反射光线，将电影摄制对于文物的影响减到最低。导演还透露，拍摄中积累的大量素材还将以纪录片的形式与观众见面。
